Glucose-6-phosphate dehydrogenase is the rate limiting enzyme in HMP shunt. Its deficiency leads to low levels of NADPH which is necessary to keep glutathione reduced which is responsible for detoxification of free radicals and peroxides. Poor defense of red blood cells against oxidizing agents as primaquine leads to hemolytic anemia.
